چکیده

Recombinant protein production in Escherichia coli is well suited to applications in the basic and applied sciences due to its associated simplicity, cost-effectiveness and the large number of genetic strategies available. However, the active-form of the marine bacterial enzyme, alginate lyase, was difficult to express in E. coli cells under standard culture conditions. In this study, we found various calcium compounds that enhanced the expression of the active enzyme. The alyPEEC gene encoding extracellular alginate lyase of Pseudoalteromonas elyakovii IAM 14594 was comprised of a 1,197 bp open reading frame encoding 398 amino acid residues with the domain G to N functioning as the mature enzyme. Three clones, pTPB24 with a 2.7 kb insert containing alyPEEC gene and paeX and their respective promoter regions, pTPB31 with a 1.6 kb insert containing only the alyPEEC gene its own promoter, and pCD11

Cloning of alginate lyase gene (alxM) and expression in Escherichia coli.

The alxM gene encoding a D-mannuronan-specific alginate lyase has been cloned from a marine bacterium isolated as an epiphyte on the brown alga, Sargassum fluitans. Expression of this gene in Escherichia coli provides a source of this enzyme for probing alginate structure and modifying the mannuronan-rich alginate polymers produced by bacterial pathogens.

Characterization of a New Cold-Adapted and Salt-Activated Polysaccharide Lyase Family 7 Alginate Lyase from Pseudoalteromonas sp. SM0524

Marine bacterial alginate lyases play a role in marine alginate degradation and carbon cycling. Although a large number of alginate lyases have been characterized, reports on alginate lyases with special characteristics are still rather less. Here, a gene alyPM encoding an alginate lyase of polysaccharide lyase family 7 (PL7) was cloned from marine Pseudoalteromonas sp. SM0524 and expressed in ...

Cloning and Expression of Heat Shock Protein 60kDa Gene from Brucella melitensis as Subunit Vaccine

Brucellosis is caused by the bacterium Brucella and affects various domestic and wild species. GroEL (Heat Shock Protein 60kDa) as one of the major antigens that stimulate the immune system, increases Brucella survival. The aim of the current study was to clone and express GroEL in Escherichia coli in order to design subunit vaccine. Amplifying was performed using specific primers. The full-len...

Sequence of a gene encoding a (poly ManA) alginate lyase active on Pseudomonas aeruginosa alginate.

The recombinant plasmid pAL-A3 bears a (poly ManA) alginate lyase-encoding gene that originates from the marine bacterium ATCC 433367 (Brown et al., Appl. Environ. Microbiol. (1991) 57, 1870-1872). The alginate lyase produced by Escherichia coli TC4 harbouring pAL-A3 was purified to protein homogeneity and the corresponding gene sequenced, giving access to the first known primary structure of a...
